什麼是 MALDI?

Matrix Assisted Laser Desorption/Ionization.

MALDI 是「基質輔助雷射脫附游離」的縮寫。
用於 MALDI 的樣品會與大量基質均勻混合。
基質吸收紫外光 (氮氣雷射,波長 337 nm) 並將其轉為熱能。一小部分的基質 (從圖中分析物頂部外層表面向下 100 nm) 會迅速 (幾個奈秒內) 加熱並和樣品一起汽化。

什麼是 TOF MS?

TOF MS 是飛行時間質譜分析 (Time of Flight Mass Spectrometry) 的縮寫。
如圖所示,會在樣品盤上產生不同大小的帶電離子。樣品盤和接地之間的電位差 V0 ,會以圖中所示方向吸引離子。受吸引離子的速率 v,會由能量守恆定律決定。由於對於所有離子,電位差 V0 都一致,具有較小 m/z 值的離子 (較輕的離子) 和帶更多電荷的離子,會更快移動通過漂移空間,直到到達偵測器為止。因此離子飛行時間會依據離子的質荷比 (m/z) 數值而異。利用此現象的質譜分析方法,稱為飛行時間質譜儀。
